Yorgo, an iGEM Safety & Security intern did a deep dive into past use of cell-free systems in iGEM. I'll chat with him more, and hope to create a list of useful devices that have worked in the past, as well as essential parts/plasmids for a cell-free collection.

Happy to contribute to the cell-free addition. The registry has a lot of interesting cell-free optimized parts; I've already recovered them during my fellowship. I can compile them and look into making a proposal!

In 2017, the Lethbridge team developed a standardized, modular, and open-source ex vivo TX-TL system: http://parts.igem.org/Collections/Cell-Free_TX-TL

In 2019, the Stanford team developed ourTXTL, a measurement standard for characterizing the relative strength of promoters in cell-free. They also implemented protocols for creating and lyophilizing cell-free lysis from E. coli and V. natriegens.
