The Readme instructions don't apply to Thunerbird Portable. You have to extract the .zip-File into Thunderbird\Data\profile. Than continue as described "Rename the extracted thunderbird-monterail folder to chrome, edit userChrome.css to choose your theme variant, and restart thunderbird. (...)"
