A few thiophene analogues were found to be active against Nsp15 in Sacchettini Lab's FRET assay.
Ten analogues and one original hit were synthesized using the route shown below:
They were tested, but none showed any activity.
By comparing the H NMR spectra of the resynthesized hit and the purchased hit from Enamine, it was discovered that the position of the nitrogen atom in the pyridine ring was different.
Further studies are ongoing to determine the exact structure.
